The Food and Drug Administration has opened applications for a new pilot program that will test whether some parts of a first-in-human drug trial application can be reviewed as they are completed rather than only after the full package is assembled. The effort could change the preparation of early clinical studies in the United States, a stage at which delays in paperwork, manufacturing information and research-site readiness can postpone testing of experimental medicines in people.
Applications for the FDA’s Expedited Investigational New Drug Pilot Program are due by 11:59 p.m. Eastern time on Oct. 30, 2026. The agency announced the launch on Sept. 15 as part of the Department of Health and Human Services’ Operation TrialBlazer initiative. The program is intended to test an administrative and regulatory process, not to establish whether a medicine works or to create a shortcut around safety review.
What the pilot changes in an IND submission
Before a drug can generally be tested in a U.S. clinical investigation, its sponsor submits an investigational new drug application, or IND, to the FDA. An IND typically includes preclinical evidence, information about how the product is made, a proposed clinical protocol and other material meant to support the initial human study.
Under the new pilot, the FDA says it may accept and review individual completed components of certain IND submissions on a rolling basis. Sponsors would work with qualified research institutions, or QRIs, that have scientific expertise relevant to preparing the application. Instead of having every element reach the agency at one moment, a sponsor could submit a component once it is complete and receive feedback earlier in the development process.
The practical goal is to find deficiencies sooner, when they may be easier to address, and to coordinate some work that can occur alongside IND preparation and agency review. The FDA said that could include, when appropriate, Institutional Review Board review and clinical-trial site activation. Institutional Review Boards independently evaluate plans for research involving people, including whether risks are reasonable and consent procedures are appropriate.
The pilot does not mean that QRIs replace the FDA or that a sponsor may begin dosing participants before the agency permits the investigation to proceed. FDA retains its authority to place a study on clinical hold, which can delay or stop a proposed trial over concerns such as inadequate safety information, manufacturing questions or problems with the study plan.
From a proposed idea to an open pilot
The current announcement marks a shift from consultation to implementation. In June, HHS announced Operation TrialBlazer and the FDA outlined a proposed expedited IND concept. The agency then sought stakeholder input, a process described at the time by Arnold & Porter’s analysis of the proposed program. The Sept. 15 FDA announcement says the pilot is now launched and accepting participation requests.
That progression helps clarify why descriptions from earlier this year referred to a prospective or exploratory model, while the agency now describes a functioning application process. The official announcement does not identify participants or report outcomes from the pilot. Independent reporting by BioPharma Dive said the FDA anticipated selecting an initial cohort of eight to 10 sponsor-QRI pairings, though that number is not stated on the agency’s public program page.
First-in-human trials are generally designed to gather early information about such issues as dosing and safety. They are not proof that an experimental treatment is effective, and a more orderly application process would not alter the scientific uncertainty that accompanies early human testing. Whether the pilot reduces delays, changes the frequency of clinical holds or improves the likelihood that a drug advances in development will depend on results that have not yet been reported.
What should not be inferred from the announcement
The FDA has framed the pilot as a test of whether rolling review and closer coordination can make the path toward a first-in-human study more predictable while preserving participant protections and scientific oversight. That is a prospective objective, not a demonstrated time saving. The agency has not said that every participating sponsor will move faster or that the initiative changes the standards used to assess an IND.
There is also a separate FDA action that should not be conflated with this pilot. In the same broader announcement, the agency said it had updated information on chemistry, manufacturing and controls, known as CMC, for Phase 1 INDs. The FDA said clearer, phase-specific CMC expectations could save six to 12 months of development time. That estimate applies to the separate CMC clarification, not to a measured result of the Expedited IND Pilot.
Manufacturing information can be particularly consequential for products entering human trials because the FDA must understand how the investigational drug will be produced and controlled well enough to protect participants. Earlier discussion of such issues, alongside preclinical and protocol questions, is the process change the pilot is designed to examine.
Industry groups have generally supported efforts to reduce avoidable administrative delays while raising implementation questions. BioPharma Dive reported that the Biotechnology Innovation Organization welcomed the potential for rolling review but questioned whether a single research institution could satisfy all of the FDA’s requirements and cautioned that the arrangement could add another layer of review. Those concerns underscore that the pilot’s usefulness will hinge not just on earlier submissions, but on how the FDA, sponsors and institutions divide responsibilities in practice.
The application deadline gives drug sponsors a near-term decision about whether to test that model. For patients, the more consequential answers will come later: whether the pilot can speed the start of well-designed studies without weakening the review intended to protect the people who enroll in them.
